okay, I have my new server (8 cpu's and 64Gb of ram) sitting in the corner I am armed with a SQL2005 Enterprise Disk, Do I have to run the install every time I want to create a new Instance?

Yes. You need to run the setup to create new instances

Remember - you also have to install SP's seperately on every instance.
